چکیده
Homogenates of pigeon heart, brain, lung, liver, and the formed elements of blood and of monkey brain, liver, spleen, kidney, lung, uterus, heart, and the formed elements of blood contain an enzyme which catalyzes the reduction of the 9-keto group of prostaglandin E to form prostaglandin F. The prostaglandin E 9-ketoreductase in all of these tissues uses NADPH much more effectively than NADH and is found in the cytoplasmic fraction. In monkey liver, there is a second prostaglandin 9-ketoreductase, which is present in the microsomal fraction and uses NADH much more efficiently than NADPH. The activities of the prostaglandin 9-ketoreductases are extremely sensitive to the oxidized cofactors, and in vivo may be regulated by the relative concentrations of reduced and oxidized coenzymes. Such enzyme activities may, in turn, regulate the diverse physiological processes involving these two classes of prostaglandins.

PGH synthase and lipoxygenase generate superoxide in the presence of NADH or NADPH.
Purified PGH synthase when acting on arachidonic acid in the presence of reduced nicotinamide-adenine dinucleotide or reduced nicotinamide-adenine dinucleotide 3'-phosphate generated superoxide in burst-like fashion. In eight experiments using different batches of enzyme, the mean +/- SE rate of superoxide generation from 100 U of enzyme measured as the superoxide dismutase-inhibitable reductio...
متن کاملVerifying of Participation of Nitric Oxide in Morphine Place Conditioning in the Rat Medial Septum Using Nicotinamide Adenine Dinucleotide Phosphate-Diaphorase (NADPH-d)
Background: Role of nitric oxide (NO) in morphine-induced conditioned place preference (CPP) has already been proposed in the rat medial septum (MS), but no molecular evidence has been provided to clear this fact. Methods: Effects of intraseptal injections of L-arginine and/or NG-nitro-L-arginine methyl ester (L-NAME) on morphine place conditioning in Wistar rats were examined. Morphine (2.5-7....
متن کاملEnhancement of DMA Chain Breakage by Bleomycin A2 in the Presence of Microsomes and Reduced Nicotinamide Adenine Dinucleotide Phosphate1
Chain breakage in DMA induced by bleomycin A (BLM) was enhanced more than 150-fold by the reduced nicotinamide adenine dinucleotide phosphate-dependent elec tron transport system of rat liver microsomes. However, the enhancement effect on DMA was partially reduced by the preincubation of BLM with the microsomal systems. BLM-Cu2' was found to stimulate considerably micro somal reduced nicotinami...
متن کاملOxidative metabolism of diethylstilbestrol by prostaglandin synthetase.
The cooxidative metabolism of the transplacental carcinogen, diethylstilbestrol (DES), was examined using ram seminal vesicle microsomes. The major extractable metabolite was beta-dienestrol (Z,Z-DIES) and represented about 35% of the added DES in 3-min incubations supplemented with arachidonic acid. Its formation was dependent upon the presence of arachidonic acid, whereas reduced nicotinamide...
متن کاملTransport of the renal carcinogen 3-hydroxymethyl-1-([3-(5-nitro-2-furyl)allydidene]amino) hydantoin by renal cortex and cooxidative metabolism by prostaglandin endoperoxide synthetase.
Transport of the renal carcinogen 3-hydroxymethyl-1-([3-(5-nitro-2-furyl)-allydidene]amino) hydantoin (HMN) by the renal cortex and metabolism by the kidney was evaluated. Organic acid and base transport by renal cortical slices was determined using [131I]Hippuran and [14C]tetraethylammonium, respectively. HMN caused a dose-dependent reversible inhibition of [131]Hippuran accumulation but did n...
